According to Greek outlet Kerkyrasimera, the bus was coming from Paleokastritsa.
But at the height of Tzavros it deviated from its course – hitting a parked truck on a side street off the main road.
The crash happened shortly after 3pm local time on Sunday.
Local media reports say that the truck driver has been seriously injured but the bus driver, aged 31, has died.
Firefighters and paramedics have also rushed to the scene.
It is understood that most passengers have been freed from the coach, but some could still be inside, Kerkyrasimera reports.
After an hour and a half operation, rescuers managed to free a mother and her child.
Those passengers needing urgent care were reportedly rushed to nearby hospitals.
Shocking video shows firemen carefully removing passengers from the bus, which appears near a concrete wall.
Corfu traffic authorities are investigating the exact cause of the crash.
Elsewhere in Greece, a Brit tourist tragically died after being pulled unconscious from the water.
The woman, 66, was swimming at Achladies beach in Skiathos and reportedly lost consciousness due to the sweltering heat.
The Brit was on holiday in Greece and went down from her hotel to the beach for a swim on Friday, CNN Greece reports.
But she was soon found by a lifeguard, who pulled her out of the water onto the shore.
The lifeguard and a doctor at the beach performed CPR on the 66-year-old as she was rushed to Skiathos Health Center, but were unable to revive her.
The woman was expected to be transferred to Volos for an autopsy.
A spokesperson for the UK Foreign Office told The Sun: “We have offered consular support to the friends of a British woman who has died in Greece.”
